The Synergistic Effect of Fotemustine and Genistein on Expressions of p53, EGFR and COX-2 Genes in Human Glioblastoma Multiforme Cell Line

Abstract: GBM is the most common primary malignant neoplasm of the central nervous system in adults. Fotemustine (FTM) is a cytotoxic alkylating agent and a lipophilic chloroethylnitrosourea derivative. Its mechanism of action consists mainly in inducing DNA strand breaks and cross-linking. Genistein, one of the soy-derived isoflavones, exerts its anticancer properties via several mechanisms, including inhibition of tyrosine phosphorylation, weak estrogenic and anti-estrogenic properties, as an antioxidant, inhibition of topoisomerase II, inhibition of angiogenesis, and induction of cell differentiation in a number of human tumors. We aimed to investigate the anti-proliferative synergistic effect of genistein with fotemustine on human glioblastoma multiforme U87-MG cells. This study was also designed to answer the following question: Do the p53, EGFR, COX-2 genes' expression patterns differ in treatment of these both drugs alone and in combination?
The Emphasis of Tumor Suppressor Genes and Oncogenes in Diagnosis and Prognosis of Anaplastic Brain Tumors

Abstract: The aim of the study is to the determine the profiles of tumor suppressor genes and oncogenes which cause brain tumor, establishing the association between the prognosis of cancer and the quantitation of genetic and epigenetic changes, and bringing a molecular approach to definite diagnosis. For this purpose, explant cell cultures are performed from the anaplastic brain tumor tissues of the cases. The expression analysis of the tumor suppressor genes (p53, RB1, PTEN, MGMT, RUNX3, DMBT1, PIKE) and oncogenes (EGFR, PIK3CA, MDM2, Olig2, GSTT1, COX–2 and hTERT) were determined by comparing the expression of GAPDH housekeeping gene using real-time online RT-PCR. The promoter regions of all the tumor suppressor genes' hypermethylation and also methylated and unmethylated copy numbers were determined with Q-PCR by using methylation specific primer and probes and the quantitation was carried out by comparing with each other. A significant difference was determined among the oncogenes; EGFR and hTERT gene expressions in patient tumor group. hTERT gene expression showed a significant difference with tumor grades. DMBT1 gene expression showed a significant difference with tumor grades. A prominent decrease was found in the aberration of tumor suppressor gene copy number in the glioma group. Gene copy number and gene expression of GSTT1 gene showed a significant correlation. RB1 and MGMT promoter methylation showed a significant difference in tumor patient group. Over expression of PIK3CA, EGFR and COX-2 among oncogenes and loss of copy number of PTEN, RB1 and RUNX3 among tumor suppressor genes found associated with short survival.
Evaluation of expression changes of genetic and epigenetic autophagy regulators in chronic myeloid leukemia cells due to epigallocatechin-3-gallate treatment

Abstract: Purpose: Chronic myeloid leukemia is characterized by the formation and activation of Bcr-Abl1 fusion tyrosine kinase. Discovery of the inhibitor which targets the fusion protein results in a significant increase in survival rates. However the development of resistance to the inhibitor necessitates determining new therapy targets. Autophagy is a remarkable dual target in recent cancer researches. It is usually aimed at inhibition of autophagy because autophagy allows cells to adapt to stress conditions. Epigallocatechin-3-gallate is a basic phytochemical found in green tea. The efficacy of epigenetic regulators, such as microRNAs, is important in the regulation of dietary recommendations as well as in treatment. In this study, it was aimed to investigate the effect of epigallocatechin-3-gallate, a flavanoid, on genetic and epigenetic factors regulating autophagy in chronic myeloid leukemia cells. Materials and methods: microRNAs which are target autophagy-related genes have been identified using appropriate databases. Expression levels of autophagy-related genes and microRNAs targeting these genes were determined using real-time quantitative PCR method in 50 micromolar Epigallocatechin-3-gallate treated K-562 cells. Results: Epigallocatechin-3-gallate treatment resulted in a significant decrease and increase the expression levels of autophagy-related genes and microRNAs targeting these genes in K-562 cells, respectively. Conclusion: In the direction of the results, epigallocatechin-3-gallate was found to cause autophagy inhibition by genetic and epigenetic regulation in chronic myeloid leukemia cells. Considering the dual mechanism of autophagy, this result suggests that epigallocatechin-3-gallate is a chemical that is important for the nutrition of patients as well as its potential to be used as a drug
The Effect of Cyclosporin A Via P53, MDM2 and PIK3CA Gene Expressions in Glioblastoma Multiforme Cells

Abstract: Objective: It is aimed to evaluate the effect of cyclosporine A in glioma cells via p53, MDM2 and PIK3CA gene expressions.Material Methods: U87MG cells are treated with 10, 30 and 60 μM CsA during 72 hour in time and dose dependent manner. The cell group that was not treated by any agent was approved of the control group. During 72 hour, total RNA was isolated from the cells which were collected every 24 hour and complementary DNA was synthesized from these cells. Dose and time dependent manner variation analysis of p53, MDM2 and PIK3CA gene expressions were performed by Real-Time Online RT-PCR. Relative analyses of these genes were achieved by rating the genes' expressions to GAPDH.Results: p53 expression showed an increase in 10μM dose of CsA in time and dose dependent manner and the rise of expression was four times at the 48th hour. MDM2 expression was increased approximately 1,5 times. When PIK3CA expression was evaluated, it was seen that, in the 24th hour, 30 μM dose of CsA showed 67% suppressed expression according to control group.Conclusions: Due to the results of the performed study by the well-known drug CsA, it is found that up-regulation of functional p53 tumor suppressor and induction of apoptosis triggered by p53 can be effective strategy in the induction of glioma cell death. These data carry a critical weight in the pharmacological explorations related with gliomas.
Uric Acid and GGT Have Causal Relations with Abdominal Obesity: A Real-Life Research in Turkish Population with 1214 Diabetics  [PDF]

Abstract: Aim: We compared the waist circumferences (WC) of 1214 diabetics with various parameters in real-life conditions. Patients and Method: The study was carried out by reviewing the anthropometric measurements and biochemical analyzes of diabetics and they were analyzed due to WC status of the patients. Results: Mean total cholesterol level was 204.6 ± 47.1 mg/dL in patients with normal WC and 211.6 ± 45.6 mg/dL in patients with increased WC (p = 0.015). Total cholesterol was found to be a risk factor for WC enlargement (OR: 1.003, 95% CI: 1.001 - 1.006, p = 0.015). HDL-cholesterol appeared to be a risk factor for the increase in WC (OR: 1.005, CI: 1.001 - 1.010, p = 0.029). Median triglyceride level was 158 (41 - 975) mg/dL in the normal WC group, while it was 176 (22 - 1379) mg/dL in the patients with increased WC (p < 0.001). Triglyceride was observed as a risk factor for WC increase (OR: 1.002, 95% CI: 1.001 - 1.003, p = 0.004). The serum uric acid level was 4.75 ± 1.6 mg/dL in the normal WC group, while it was 5.09 ± 1.5 mg/dL in the increased WC group (p = 0.002). Serum uric acid level was appeared as a risk factor for increased WC (OR: 1.162, 95% CI: 1.057 - 1.276, p = 0.002). The median serum GGT level was 27 (7 - 174) mg/dL in WC normal group and 28 (8 - 730) mg/dL in increased WC diabetics (p = 0.029). Serum GGT level was a risk factor for increased WC (OR: 1.162, 95% CI: 1.001 - 1.010, p = 0.029). Conclusion: We have demonstrated that the measurement of WC has an important role in diabetes management and is related with inflammatory parameters such as uric acid and GGT.

Using Concept Maps as a Method of Assessment in Work-Energy Subject

Abstract: The purpose of this study is to analyze the concepts of work-energy subject in 7th grade science course and the relations among them. Concept maps were used as a method of assessment and relationships between concepts of work-energy topic were examined using this way. The research was performed by contribution of 50 students within a period of 7 weeks. Each student formed his/her concept map related to the topic. These maps were assessed according to Novak`s score system. The assessment was made with two raters and g coefficient was found as 0.97. Additionally work-energy achievement test was applied at the end of this process. Results obtained from the concept maps and the results of the test were compared. As a result, the correlation between the two assessment methods was computed as 0.416. The relationships between the concepts were examined deeply. Serious defects were determined in the linking words in the concept maps of the students.

Sleep Apnea Detection Using Adaptive Neuro Fuzzy Inference System  [PDF]

Abstract:

This paper presents an efficient and easy implemented method for detecting minute based analysis of sleep apnea. The nasal, chest and abdominal based respiratory signals extracted from polysomnography recordings are obtained from PhysioNet apnea-ECG database. Wavelet transforms are applied on the 1-minute and 3-minute length recordings. According to the preliminary tests, the variances of 10th and 11th detail components can be used as discriminative features for apneas. The features obtained from total 8 recordings are used for training and testing of an adaptive neuro fuzzy inference system (ANFIS). Training and testing process have been repeated by using the randomly obtained five different sequences of whole data for generalization of the ANFIS. According to results, ANFIS based classification has sufficient accuracy for apnea detection considering of each type of respiratory. However, the best result is obtained by analyzing the 3-minute length nasal based respiratory signal. In this study, classification accuracies have been obtained greater than 95.2% for each of the five sequences of entire data.

Parasitoid Complex and New Host Plants of the Gypsy Moth, Lymantria dispar L. in the Lakes District, Turkey

Abstract: The present study determined egg, larval and pupal parasitoids of the gypsy moth Lymantria dispar L. (Lepidoptera: Lymantriidae), which is an important forest pest in Turkey. High populations of the gypsy moth have been observed recently in oak forests and coniferous forests in the Lakes District of Turkey. Pinus brutia Ten and Cedrus libani A. Rich are reported for the 1st time in the world as host plants of L. dispar. Its parasitoid complex and impact were analyzed in a 2-years study. In larval and pupal stages of the pest, the tachinids Exorista segregata (Rond.), Compsilura concinnata Meigen, Drino inconspicua Meigen and Pales pavida Meigen, the chalcid Brachymeria intermedia (Nees), the torymid Monodontomerus aereus Walk. The braconid Apanteles xanthostigma (Haliday) and the egg parasitoids Ooencrytus kuvanae (Howard) (Hym. Encyrtidae), Anastatus disparis Rusch and Anastatus bifasciatus (Fonsc.) (Hym: Eupelmidae) were recorded. O. kuvanae had the greatest impact on L. dispar populations. A. xanthostigma is a new record for Turkey and also a new record for the world as a parasitoid of L. dispar.
